We are seeking a highly motivated and innovative PhD-level scientist to join the Target Discovery Group in Pfizer's Oncology Research &; Development organization. The successful applicant will employ multiple genomic and functional genomic platforms to identify and validate novel small molecule or antibody drug targets. The primary therapeutic focus of this position will be leading programs to define novel immune-oncology targets but the successful candidate will be expected to be additionally capable of contributing to research programs to identify synthetic lethal oncology targets arising from tumor genetic vulnerabilities.
The successful candidate will work independently as part of a research team that is focused on identifying and validating novel cancer drug targets. The candidate is expected to utilize expertise in the fields of molecular and cellular biology, cancer biology, immunology and functional genomics to design, execute and interpret experiments and intellectually drive programs to develop novel oncology targets for therapeutic intervention.
Specific technical duties include:
Utilize molecular and cellular biology skills including DNA cloning, cell culture, lentiviral shRNA and CRISPR knockdown technologies followed by global MiSeq analysis to execute high-throughput genomic screens.
Apply both genetic and pharmacologic approaches to modulate target candidates in vitro to assess the functional impact on both immune and cancer cell biology. These include RNAi, CRISPR, small molecules, antibodies and other pharmacologic approaches to perturb gene targets in vitro and develop molecular, cellular and phenotypic assays for functional analysis.
Establish and execute molecular and cell-based assays across a wide variety of technical platforms (qPCR, Western blotting, FACS, immunofluorescence, ELISA, HCS etc.) to assess target modulation and its impact on immune function and tumor-susceptibility, cell viability and cell cycle disruption, transcription and downstream signal transduction.
Utilize expertise in cancer biology to design, execute, and analyze experiments to advance mechanism of action target biology and develop innovative approaches for targeting cancer. Coordinate with In Vivo Pharmacology group to validate target candidates in animal models.
Utilize electronic data capture/analysis tools such as Excel, GraphPad, Biobook, Spotfire and PowerPoint for experimental documentation and data analysis/presentations
Present experimental results at both internal group meetings in the Oncology Research &; Development organization and through external meetings and publications.
PhD in Cancer Biology, Immunology, Cell Biology (or a comparable biological science) plus post-doctoral training or equivalent experience in cancer biology, immunology, cell signaling or a related discipline. Well-qualified candidates will have a track record of scientific accomplishments as evidenced by a strong publication record.
Minimum of 2 years of experience in academia or industry focused on immune and/or cancer biology and drug discovery analysis.
Deep knowledge of literature in areas of both immune and tumor cell biology in addition to strong motivation and creativity required to drive drug discovery target programs.
Hands on experience in in multiple molecular and cellar biology techniques including genetic manipulation, generation of engineered cell lines, high-throughput assays, flow cytometry, phenotypic assays are required.
Backgrounds encompassing immunooncology and/or experience in target identification/validation is a plus.
Strong interpersonal skills and excellent communication abilities are required for this role.
Technical Skill Requirements:
Technical knowledge of cancer biology including oncogenic signaling pathways and/or expertise in immune cell biology is required.
Expertise in molecular biology, mammalian cell culture techniques and cell-based functional assays is required.
Expertise in gene knockdown technologies utilizing RNAi and CRISPR platforms, gene expression analysis (qPCR, Western blotting, FACS analysis) and cell-based phenotypic assays is critical.
Strong track record in screening approaches for target identification, utilization of inhibitors or genetic manipulations for target validation and/or expertise in immune-oncology is highly valued.
Excellent verbal/written communication and interpersonal skills with an emphasis on working in a team environment to accomplish impactful results.
Ability to work in a fast paced dynamic environment. Self-motivation and demonstrated capability to independently drive research programs.
