The Rare Diseases Research Unit (RDRU) of Pfizer is committed to the development of new therapies to address the unmet medical needs of those individuals with rare and neglected diseases. Recently, the RDRU established Bamboo Therapeutics in Chapel Hill, North Carolina in order to develop novel gene therapies using AAV.
The RDRU is seeking to recruit a laboratory scientist to support Bamboo's goal of advancing development of novel capsid for targeted gene transfer, immune response, and efficient and persistent tissue specific transgene expression. The position requires a strong background in viral immunology in general and experience in recombinant AAV vector immune epitope characterization.
The successful candidate will contribute directly to projects in the research group pertaining to testing and optimization of transgene expression and persistence after AAV vector administration in vivo in mouse and non-human primate models. The candidate will be expected to develop structural predictors of rAAV immunogenicity and stability.
The candidate will contribute to capsid development projects emphasizing structural analysis of AAV capsid interactions with antibodies and immune cells, and activation of innate immune responses. This position involves working closely with a multidisciplinary project team to design and execute multi-day experiments in addition to performing complex data analysis. The candidate must be a strong experimentalist with the ability to lead from the bench while playing a key role in hypothesis generation. Our lab develops emerging targets applicable to Duchenne Muscular Dystrophy and other rare diseases rooted in human biology and genetics. We are seeking new members of our team that focuses on innovative solutions to the challenges of rare disease research sphere. These scientist positions will focus primarily on the use of AAV.
The key responsibilities for this role are to perform thorough experiment planning, execution, and analysis of structural features of the AAV capsid that impact the immune responses to vector administration in mouse, large animal, and non-human primate models. This position will focus on altering and minimizing the immunogenicity of the AAV capsid and it's interactions with pre-existing anti-AAV antibodies. Critical to the success of this position is a fundamental understanding of viral immunology in general and specific host responses to viral vector and transgene antigens.
PhD in viral immunology with 5+ years of laboratory experience including degree research
Candidate must have 1+ years experience working with AAV immunology.
Candidates must have 2+ years of independent lab research experience working with immune assay development.
Candidate must have experience working with a research team focused on project-specific goals as well as able to facilitate joint collaboration between research teams.
Expertise in publications, presentations, and managing subordinate research projects is necessary.
Skills and attributes:
Skills in analysis and prediction of structural features of the AAV capsid that impact immunogenicity and stability
Track record of hypothesis-driven research that employs complex immunological analysis
Experience with the development of specific assays to study host response to vector transduction and persistence.
Team player who can communicate effectively in written and spoken English.
Proficiency in Excel, Word, PowerPoint and scientific graphing software. Experience with statistical analysis software such as R and ability to perform basic scripting programming is desirable. Experience with PyMol and other structural programs for analyzing 3D data are required.
